Start in Cairns, follow the coast through Port Douglas and cross the Daintree River to Cape Tribulation, then turn inland for the Atherton Tablelands. Mission Beach brings the journey back to the sea before the drive returns to Cairns. A Great Barrier Reef trip is an extra day to arrange with a boat operator; it is not included in this road loop.
